Transgenic Microalgae Expressing Double-Stranded RNA as Potential Feed Supplements for Controlling White Spot Syndrome in Shrimp Aquaculture

چکیده

Viral infection of farmed fish and shellfish represents a major issue within the aquaculture industry. One potential control strategy involves RNA interference viral gene expression through oral delivery specific double-stranded (dsRNA). In previous work, we have shown that recombinant dsRNA can be produced in chloroplast edible microalga Chlamydomonas reinhardtii used to disease shrimp. Here, report significant improvement antiviral production its use protect shrimp against white spot syndrome virus (WSSV). A new for synthesis was developed uses two convergent copies endogenous rrnS promoter drive high-level transcription both strands WSSV element chloroplast. Quantitative RT-PCR indicated ~119 ng per liter culture transgenic microalga. This an ~10-fold increase relative our report. The engineered alga assessed ability prevent when fed larvae prior challenge with virus. survival given feed supplemented dried containing significantly enhanced (~69% survival) negative (<10% survival). findings suggest this platform could employed as low-cost, low-tech method aquaculture.

Design and Evaluation of a PCR Method for Detecting White Spot Syndrome Virus in Shrimp

Background: White spot syndrome virus (wssv) is the causing agent for white spot disease in shrimp and many crustaceans. This disease is highly contagious and can cause death within 3–10 days under normal culture conditions. Therefore, early diagnosis of the virus is a necessity. Materials and Methods: Primers were designed for three regions of the virus genome and one region of the shrimp geno...

Large-scale production and antiviral efficacy of multi-target double-stranded RNA for the prevention of white spot syndrome virus (WSSV) in shrimp

BACKGROUND RNA interference (RNAi) is a specific and effective approach for inhibiting viral replication by introducing double-stranded (ds)RNA targeting the viral gene. In this study, we employed a combinatorial approach to interfere multiple gene functions of white spot syndrome virus (WSSV), the most lethal shrimp virus, using a single-batch of dsRNA, so-called "multi-WSSV dsRNA." A co-culti...

Enhanced Whitefly Resistance in Transgenic Tobacco Plants Expressing Double Stranded RNA of v-ATPase A Gene

BACKGROUND Expression of double strand RNA (dsRNA) designed against important insect genes in transgenic plants have been shown to give protection against pests through RNA interference (RNAi), thus opening the way for a new generation of insect-resistant crops. We have earlier compared the efficacy of dsRNAs/siRNAs, against a number of target genes, for interference in growth of whitefly (Bemi...

Fish processing wastes used as feed ingredient for animal feed and aquaculture feed

Fish wastes management has become a global problem from the last years. Dispose of seafood wastes cause environmental pollution. To overcome this issue these unwanted seafood products are used for the formation of animal feed and aquaculture feed. These unwanted products include small fish and those parts of fish which are not used as human food. These unwanted parts include viscera, head, fins...
